The principle of a hot air oven revolves around the process of dry air sterilization using convection, conduction, and radiation. The oven contains heating elements that generate heat, which is then transferred to the air inside the chamber. The air circulation is facilitated by fans to ensure even distribution of the heated air throughout the oven.

When items or samples are placed inside the oven, their surfaces come into contact with the hot and dry air. The heat energy is transferred from the air to the external surface of the items through the process of conduction. As the heat continues to penetrate the items, it gradually moves toward their center, reaching deeper layers through the same conduction process.

Microorganisms present on the surfaces of the items are particularly affected by the hot air. The heat causes the water content inside the microorganisms to evaporate. This process of evaporation leads to several detrimental effects on the microorganisms. Firstly, the oxidative damage of cellular constituents occurs, as the heat promotes chemical reactions that generate reactive oxygen species, which can harm the microorganism’s cellular components.

·         A hot air oven, also known as a forced air circulating oven, is a laboratory instrument used for the sterilization of laboratory equipment and other materials using dry heat. It is particularly suitable for sterilizing items that cannot be exposed to moisture or materials that are not prone to melting, catching fire, or changing form when subjected to high temperatures.

·         The main purpose of a hot air oven is to provide a controlled environment where high temperatures can be maintained consistently. The oven achieves this through the circulation of air, ensuring even distribution of heat throughout the chamber.

·         While there are certain materials that are not suitable for sterilization in a hot air oven, such as surgical dressings, rubber items, and certain plastics, there are several items that can be effectively sterilized. These include glassware like petri dishes, flasks, pipettes, and test tubes, powders like starch, zinc oxide, and sulfadiazine, materials containing oils, and metal equipment like scalpels, scissors, and blades.

·         Hot air ovens employ extremely high temperatures over extended periods to eliminate microorganisms and bacterial spores. Commonly used temperature-time relationships for sterilization in hot air ovens include 170 degrees Celsius for 30 minutes, 160 degrees Celsius for 60 minutes, and 150 degrees Celsius for 150 minutes.

·         Due to its straightforward standard operating procedure and cost-effectiveness, hot air ovens are widely utilized in medical industries for the sterilization of laboratory instruments and materials. Additionally, hot air ovens facilitate rapid drying processes.

·         The concept of dry heat sterilization using a hot air oven was initially developed by the renowned scientist Louis Pasteur. The temperature range of a hot air oven typically spans from 50 to 300 degrees Celsius, and the temperature can be regulated using a temperature regulator.

·         The forced air circulation within the oven ensures uniform temperature distribution, thereby ensuring consistent sterilization throughout the chamber. In the hot air oven, the surface of the material is first sterilized, followed by a gradual penetration of heat into the center of the item.
